BACKGROUND: Tie1 orphan receptor has become a focus of research, Tie1 can form a polymer with Tie2, regulate the Ang/Tie2 pathway and play a vital role in pathological angiogenesis and tumor progression, the function of Tie1 has remained uncertain in the progression of cervical cancer (CC). Here, we investigated the functional influences of Tie1 overexpress on CC in vitro and in vivo. METHODS: We used Immunohistochemistry (IHC) analysis to detect the relative expression of Tie1 in CC, and we analyzed its connection with the overall survival (OS) and progression free survival (PFS)of CC patients. To prove the role of Tie1 in cell proliferation and metastatic, Tie1 expression in CC cell lines was upregulated by lentivirus. RESULTS: The high expression of Tie1 in tumor cells of cervical cancer tissues is significantly correlated with FIGO stage, differentiated tumors, tumors with diameters, deep stromal invasion. We found that cell progression was promoted in Tie1-overexpress CC cell lines in vivo and in vitro. Tie1 potentially exerts a commanding influence on the expression of markers associated with epithelial-mesenchymal transition (EMT) and the PI3K/AKT signaling pathway. CONCLUSIONS: Our research indicates that Tie1 is highly connected to CC progression as it may play a role in the EMT process through the PI3K/AKT signaling pathway.

LAMA5 (laminin α5) is a member of the laminin family. Despite the recent research implicating LAMA5 in cancer, the function of LAMA5 has remained uncertain in the progression of ovarian cancer (OC). Here, we investigated the functional influences of LAMA5 knockdown on OC in vitro and in vivo. In this study, we used immunohistochemistry (IHC) analysis to detect the relative expression of LAMA5 in OC and non-cancer tissues, and we analyzed its connection with the overall survival (OS) of OC patients. To prove the role of LAMA5 in cell proliferation, migration, and invasion, LAMA5 expression in OC cell lines was inhibited by lentivirus. Compared with normal fallopian tube tissue, epithelial ovarian cancer (EOC) tissue showed critically higher LAMA5 expression levels; additionally, high LAMA5 levels were a poor predictor of OS. We found that cell progression was restrained in LAMA5-knockdown OC cell lines in vivo and in vitro. Finally, LAMA5 might be a commanding inducer of the expression of epithelial-mesenchymal transition (EMT) and Notch signaling pathway-related markers. Together, our research indicates that LAMA5 is highly connected to OC progression as it may play a role in the EMT process through the Notch signaling pathway.

BACKGROUND: Ovarian cancer (OC) is one of the most common malignancies in women. It has a poor prognosis owing to its recurrence and metastasis. Unfortunately, reliable markers for early diagnosis and prognosis of OC are lacking. Our research aimed to investigate the value of the six-transmembrane epithelial antigen of prostate family member 3 (STEAP3) as a prognostic predictor and therapeutic target in OC using bioinformatics analysis. METHODS: STEAP3 expression and clinical data were acquired from The Cancer Genome Atlas (TCGA), Genotype-Tissue Expression (GTEx), and Gene Expression Omnibus (GEO). Unsupervised clustering was used to identify molecular subtypes. Prognosis, tumor immune microenvironment (TIME), stemness indexes, and functional enrichment analysis were compared between two definite clusters. Through the least absolute shrinkage and selection operator (LASSO) regression analysis, a STEAP3-based risk model was developed, and the predictive effectiveness of this signature was confirmed using GEO datasets. A nomogram was used to predict the survival possibility of patients. Additionally, TIME, tumor immune dysfunction and exclusion (TIDE), stemness indexes, somatic mutations, and drug sensitivity were evaluated in different risk groups with OC. STEAP3 protein expression was detected using immunohistochemistry (IHC). RESULTS: STEAP3 displayed marked overexpression in OC. STEAP3 is an independent risk factor for OC. Based on the mRNA levels of STEAP3-related genes (SRGs), two distinct clusters were identified. Patients in the cluster 2 (C2) subgroup had a considerably worse prognosis, higher immune cell infiltration, and lower stemness scores. Pathways involved in tumorigenesis and immunity were highly enriched in the C2 subgroup. A prognostic model based on 13 SRGs was further developed. Kaplan-Meier analysis indicated that the overall survival (OS) of high-risk patients was poor. The risk score was significantly associated with TIME, TIDE, stemness indexes, tumor mutation burden (TMB), immunotherapy response, and drug sensitivity. Finally, IHC revealed that STEAP3 protein expression was noticeably elevated in OC, and overexpression of STEAP3 predicted poor OS and relapse-free survival (RFS) of patients. CONCLUSION: In summary, this study revealed that STEAP3 reliably predicts patient prognosis and provides novel ideas for OC immunotherapy.

The effect of the nano-sized γ' phase on the ultrasonic and mechanical properties of the IN939 superalloy was investigated. The results indicate that the microstructure characteristics of the nano-sized γ' phase directly affected the ultrasonic longitudinal velocity, the attenuation coefficient, and the mechanical properties. The ultrasonic longitudinal velocity increased with the volume fraction of the γ' phase, whereas the attenuation coefficient was similar to the fractional change in the γ channel width. The lower fractional change in the γ channel width, in combination with a high volume fraction of the γ' phase, was conducive to improving the mechanical properties of the superalloy. Additionally, the variation in the ultrasonic properties could reflect the variation in the mechanical properties of the IN939 superalloy, which was beneficial for optimizing the heat treatment process and characterizing the γ' phase precipitation behavior in a nondestructive manner.

Surfactant-enhanced remediation is less applicable for the treatment of dichlorobenzene (DCB)-contaminated soil. In this study, water solubility enhancements of o-dichlorobenzene (o-DCB) and p-dichlorobenzene (p-DCB) by micellar solutions of biosurfactants (saponin, alkyl polyglycoside) and chemically synthetic surfactant (Tween 80) were measured and compared. Solubilities of o,p-DCB in water were greatly enhanced in a linear fashion by each of Tween 80, saponin, and alkyl polyglycoside. Solubility enhancement efficiencies of surfactants followed the order of Tween 80 > saponin > alkyl polyglycoside. However, the ex situ soil washing experiment demonstrated the opposite result. The removal efficiency of o,p-DCB by biosurfactant saponin and alkyl polyglycoside was higher than that of chemically synthetic surfactant Tween 80 in contaminated soil. This difference may be due to the different adsorption behaviors of the surfactants onto soil. In addition, elution kinetics for o,p-DCB were relatively fast, with apparent elution equilibrium reached within 360 min, and can be described by a pseudo first-order kinetic equation. The elution process of o,p-DCB in soil-aqueous systems obeyed four-parameter biphasic first-order kinetic model including rapid and slow phases. The results confirmed potential application of saponin and alkyl polyglycoside in elution solution for enhanced remediation of DCB-contaminated soil.

A single-mode Brillouin-erbium fiber laser (BEFL) is realized using only a 5 m single-mode fiber (SMF) as Brillouin gain medium, which to our best knowledge is the shortest SMF ever used to construct a BEFL. The Brillouin pump (BP) preamplification technique is adopted using a bidirectionally amplified erbium-doped fiber amplifier inside the BEFL cavity. This BEFL presents a high optical signal-to-noise ratio (~40 dB), a large output power (~10 mW), and a low 980 nm pump threshold (~15 mW). Experimental results indicate that this threshold increases with the BP power. Two lower and upper BP thresholds exist for the BEFL onset and turnoff, respectively, for a fixed 980 nm pump power. Moreover, the output power of this BEFL increases linearly with the 980 nm pump power at the same slope efficiency of ~10% for various BP powers, but decreases linearly with the BP power at the same slope efficiency of ~-26% for various 980 nm pump powers.
